The present northern wing of the hotel building was the site of a fort, a small fortress built in 1829 for defense against sea attacks. During the expansion of the fort in 1843, a mysterious monolith inscribed with unknown inscriptions, the so-called “Singapore Stone” (the remains of which now constitute a national treasure of the state) was found here, but unfortunately blown up, testifying to the importance of this place even before the British. Fort Fullerton served as Singapore's first post office and first stock exchange.

In 1920, plans for a new building appeared. Funds were raised by 1924 (totaling over $4 million) and construction was completed in 1928.

The building opened on June 27, 1928, and the governor officially named it the Fullerton Building after his predecessor. The building housed the Post Office, the Stock Exchange, the Singapore Club, the Maritime Department, the Import and Export Department and several other government departments. One of the features of the building was the lighthouse crowning it.
The closest thing to the building's current function was the Singapore Club, which was housed in the building until 1961, an exclusive elite community that settled on the upper floors to provide its members with fulfillment and comfort. In the Club rooms, members dined, relaxed, socialized, played billiards and cards. Bedrooms in the attic floors could accommodate guests.

During World War 2, a British hospital operated in the building. After the Japanese occupation, it housed the headquarters of the Japanese Military Administration of Singapore. In the post-war period, the building continued to be an administrative building until 1997, when a Hong Kong company bought it to convert it into a first-class hotel.

A full renovation was completed by the end of 2000 and the historic building entered the new millennium as a 400-bed upper category hotel.

In 2001, The Fullerton Hotel received an architectural award from the local government for the restoration and enhancement of Singapore's architectural heritage, the former Fullerton Building.
